    摘要:目的〓探讨miR-103a-3p在人肺癌组织中的表达及对肺癌A549细胞增殖、迁移和侵袭的影响。 方法:收集南通市肿瘤医院52例肺癌患者手术组织标本,qRT-PCR检测肺癌组织及癌旁组织中miR-103a-3p的表达;采用Lipofecta mineTM 3000 将miR-103a-3p mimic、mimic NC、miR-103a-3p inhibitor及inhibitor NC转染至A549细胞中,qRT-PCR检测miR-103a-3p的转染效率;CCK8实验检测其细胞增殖能力;流式细胞术检测其细胞凋亡能力;划痕愈合实验检测细胞的迁移能力;Transwell小室实验检测细胞的侵袭能力。 结果:miR-103a-3p在肺癌组织中的表达水平明显低于癌旁组织(p<0.01);qRT-PCR结果显示,转染miR-103a-3p mimic组细胞中miR-103a-3p的表达水平明显高于mimic NC组,且细胞增殖、迁移和侵袭能力受到抑制;而转染miR-103a-3p inhibitor 组细胞中miR-103a-3p的表达水平低于inhibitor NC组,且细胞增殖、迁移和侵袭能力增强;但细胞凋亡在4组中变化均不明显。 结论:miR-103a-3p在肺癌组织中低表达,miR-103a-3p能够抑制肺癌A549细胞的增殖、迁移和侵袭。

    Abstract:

    Abstract:Objective: To investigate the expression of miRNA-103a-3p (miR-103a-3p) in lung cancer tissues and its effect on proliferation, migration and invasion of lung cancer A549 cells. Methods: 52 tissues of lung cancer patients were collected from Nantong tumor hospital. The expressions of miR-103a-3p were compared between lung cancer tissues and the para-carcinoma tissues by real-time PCR. miR-103a-3p mimic, mimic NC, miR-103a-3p inhibitor and inhibitor NC were transfected into lung cancer 549 cells by Lipofecta mineTM 3000 and the transfection efficiency was detected by QRT-PCR. The cell proliferation was evaluated by CCK8 assay. The cell apoptosis was detected by flow cytometry assay. The cell migration and invasion abilities were detected by wound healing assay and transwell assay. Results: The expressions of miR-103a-3p in lung cancer tissues were less than those in para-carcinoma tissues (p<0.01). The expressions of miR-103a-3p in miR-103a-3p mimic group were higher than in mimic NC group, and the proliferation, migration and invasion abilities of A549 cells were decreased. While the expressions of miR-103a-3p in miR-103a-3p inhibitor group were less than inhibitor NC group, and the proliferation, migration and invasion abilities of A549 cells were increased. But the cell apoptosis had no significant difference in four groups. Conclusion: miR-103a-3p is down-regulated in lung cancer tissues. The proliferation, migration and invasion abilities of lung cancer A549 cells were inhibited after the transfection of miR-103a-3p.
