    摘要:目的探讨血浆miRNA水平与慢性阻塞性肺疾病(COPD)严重程度的相关性。方法利用荧 光定量PCR( qPCR )检测健康人群( 10例)及COPD患者( 10例)血浆miRNA的表达水平,筛选与COPD患者密切相关的miRNA。进一步在健康人群( 28例)和COPD慢性阻塞性肺疾病全球创议(GOLD) I级(30 例)、I级(32例)、I级(30例)及IV级(28例)患者中明确候选miRNA单独及联合检测对COPD严重程度的预测价值。结果COPD患者血浆miR-320c, miR-126 及miR-34c水平显著高于健康人群(P<0.01)。随着COPD级别的增加,血浆miR-320c、miR-126及miR-34c 的表达水平亦增加。miR-320c 、miR-126、miR-34c单独及联合检测预测中重度COPD患者的ROC曲线下面积( AUCMC)分别为0.822、0.705、0.783及0.892。结论血浆miR-320c.miR-126及miR-34c可预测COPD严重程度,其联合检测的预测效能显著高于单项miRNA。

    Abstract:

    Abstract : Objective To investigate the correlation between plasma miRNA levels and the seventy of chronic obstructive pulmonary disease( COPD). Methods Quantitative fluorescent PCR( qPCR) was employed to analyze plasma miRNAs in a cohort comprising 10 healthy subjects and 10 COPD patients. Subsequently, miRNAs closely associated with COPD were identified. Additionally ,plasma samples from healthy adults( n= 28),COPD patients with GOLD grade I(n=30), grade I(n=32), grade川(n=30), and grade IV (n=28) were utilized to select candidate miRNAs. The roles of these candidate miRNAs, both individually and in combination, in predicting COPD severity were validated. Results The levels of plasma miR-320c, miR-126 , and miR-34c were found to be significantly elevated in COPD patients compared to healthy individuals( P<0.01). Furthermore, the expression abundance of these miRNAs showed an increasing trend with higher COPD grades. The areas under the receiver operating characteristie( ROC) curve of miR-320c,miR-126, miR-34c, and their combination for predicting COPD severity were 0.822, 0.705 , 0.783, and 0.892, respectively. Conclusion The levels of plasma miR-320c,miR-126,and miR 34c can predict the severity of COPD, and their combination detection exhibits significantly higher predictive ffcacy than single miRNA analysis.
